What are your rights if you inherit an estate under condition you look after a sibling?

I agree with you tell the mother you are not interested in anything if thats the way things have to be. Seems to me like she wants to make sure he still has a free ride once she is gone. Tell her thanks but no thanks. If she puts it that way in the will then you would have to follow that.
